Les piARNs, en première ligne pour la défense des koalas contre l’invasion rétrovirale

A la fin des années 1990, l’augmentation des cas de leucémies, lymphomes, infections par Chlamydiae chez les populations de koalas sauvages et en captivité a été associée à l’infection par des gamma rétrovirus. Cette contamination a sans doute pour origine une transmission inter-espèces à partir de rétrovirus de rongeurs. En Australie, cette infection a progressé du nord au sud pour atteindre la quasi-totalité du territoire. Seules les populations de koala des îles du sud ne sont pas encore infectées à 100%. Le KoRV-A a pour caractéristique d’infecter certaines cellules somatiques, mais également les cellules germinales, ce qui fait de cette infection un modèle pour l’observation de l’invasion d’un génome par un rétrovirus qui devient endogène. Dans un article publié dans Cell le 17 octobre 2019, Tianxong Yu et coll analysent le génome de koalas infectés et décrivent un mécanisme inné de défense de l’intégrité du génome cellulaire. Les PIWI-interacting (piARNs) sont des petits ARN de 21 à 35 nucléotides qui ont été décrits chez quasiment toutes les espèces et qui ont un rôle important dans la protection du génome des lignées germinales contre l’invasion et la déstabilisation par des éléments transposables. Ces piARNs inactivent les éléments transposables en s’appariant par complémentarité de séquence avec les ARNm des rétrotransposons et en les guidant vers les protéines PIWI pour être dégradés par clivage au niveau du 10e nucléotide d’appariement. Lors de ce clivage, un piARN secondaire est créé, qui peut lui-même cibler les transcrits provenant des clusters de gènes codant pour les piARNs, ce qui permet d’amplifier l’action des piARNs par un cycle dit de « ping-pong ».

En comparant le génome de 4 animaux au génome de koala référence, les auteurs de l’article montrent d’abord que le KoRV-A, mais également 3 autres rétrovirus endogènes, sont actifs et exprimés dans différents tissus, y compris les tissus germinaux. En effet, chacun des animaux présentait environ 60 nouvelles insertions de KORV-A par rapport au génome de référence. L’analyse de l’expression de ces rétrotransposons et du taux de transpositions aléatoires suggère cependant une activité moindre des rétrovirus dans les cellules germinales.

Le mode d’action habituel des piARNs est basé sur une complémentarité de séquence entre les piARNs antisens et les ARNm cibles. Ce mécanisme correspond à une réponse adaptative et spécifique. De façon inattendue, les auteurs décrivent ici l’accumulation de piARNs sens, dérivés du clivage de transcrits non épissés produits à partir de l’ADN proviral du KoRV-A, dans les tissus germinaux des koalas. Le ratio entre transcrits épissés/transcrits non épissés est plus élevé dans les cellules germinales que dans les cellules somatiques, suggérant une dégradation préférentielle des transcrits non épissés dans ces cellules. Les auteurs suggèrent que cette étape repose sur la reconnaissance innée de séquences d’origine rétrovirale, dont le mécanisme reste encore à élucider.
