Les protéines Fe-S sont des protéines ubiquitaires qui se développent dans les cellules de l’organisme. Elles sont omniprésentes et contrôlent une grande variété de processus biologiques clés ; par conséquent, le maintien de l’homéostasie des grappes Fe-S est une tâche essentielle pour tous les organismes. Cet article met alors en lumière la diversité des mécanismes homéostatiques exploités par les bactéries pour utiliser de manière coordonnée deux machineries de biogénèse des grappes Fe-S.

Ici, il est présenté un exemple de la manière dont une bactérie de la branche des Deltaproteobacteria coordonne l’expression de deux machineries de biogénèse des grappes Fe-S. Les résultats révèlent ainsi un nouveau modèle de coordination de la biogénèse des grappes Fe-S, soulignant les caractéristiques uniques et communes qui ont émergé indépendamment chez des bactéries phylogénétiquement éloignées pour maintenir l’homéostasie des clusters Fe-S en réponse à des changements environnementaux. La régulation est orchestrée par un régulateur transcriptionnel non caractérisé auparavant, RisR, appartenant à la superfamille Rrf2, dont les membres sont connus pour détecter divers stress environnementaux fréquemment rencontrés par les bactéries. Comprendre comment M. xanthus maintient l’homéostasie des amas Fe-S via la régulation RisR a révélé une stratégie reflétant le mode de vie aérobie de cet organisme. Ces nouvelles connaissances ouvrent également la voie à l’amélioration de la production de métabolites secondaires Fe-S-dépendants en utilisant M. xanthus comme châssis.

Myxococcus xanthus possède en effet deux machineries de biogenèse des clusters Fe-S, ISC (cluster fer-soufre) et SUF (mobilisation du soufre). Il est montré dans cet article que par rapport aux entérobactéries phylogénétiquement éloignées, qui possèdent également les deux machineries, M. xanthus a développé un schéma transcriptionnel indépendant pour réguler de manière coordonnée l’expression de ces machineries. Cette réponse transcriptionnelle est dirigée par RisR, qui appartient à un sous-groupe phylogénétiquement distant et biochimiquement distinct de la famille des facteurs de transcription Rrf2, en comparaison avec IscR qui régule les opérons isc et suf chez les entérobactéries. Ainsi, RisR héberge un cluster Fe-S et holo-RisR agit comme un répresseur des opérons isc et suf, contrairement à Escherichia coli, où holo-IscR réprime l’opéron isc tandis que apo-IscR active l’opéron suf. Cet article établit que la nature du cluster et les sites de liaison à l’ADN de RisR, dans les opérons isc et suf, divergent de ceux d’IscR. Egalement, chez M. xanthus, les deux machineries semblent être totalement interchangeables dans le maintien des niveaux de biogénèse des clusters Fe-S et dans la synthèse du cluster Fe-S pour leur régulateur commun, RisR. De plus, en réponse au stress oxydatif et à la limitation en fer, l’augmentation transcriptionnelle des opérons isc et suf de M. xanthus était uniquement médiée par RisR et la contribution de la machinerie SUF était plus importante que celle de la machinerie ISC.

Etant donné que les protéines Fe-S sont devenues des enzymes clés dans la production/modification de métabolites et en considérant le besoin croissant de nouveaux antimicrobiens, cette étude ouvre la voie à l’amélioration de la biosynthèse à l’échelle industrielle de molécules dépendant de la grappe Fe-S chez M. xanthus
