Post-doc (deux ans) : Interactions moléculaires de Mycobacterium bovis et Mycobacterium avium ssp. paratuberculose avec des amibes environnementales (Poitiers, France)

Les amibes libres de l’environnement pourraient être des réservoirs et des vecteurs de Mycobacterium bovis (17) et/ou de M. avium ssp. paratuberculosis (Map) et/ou d’autres mycobactéries qui interfèrent avec le diagnostic de tuberculose. Elles pourraient jouer un rôle important dans la persistance de la tuberculose bovine et de la paratuberculose respectivement. En effet, il a été démontré expérimentalement que certaines mycobactéries peuvent persister ou se multiplier au sein des amibes in vitro, mais très peu d’études ont abordé le rôle du amibes environnementales dans la survie et la virulence des mycobactéries. Les objectifs du projet sont 1) d’étudier la permissivité des isolats environnementaux d’amibes à l’infection par Mbo et Map in vitro et au sein des mésocosmes et 2) d’étudier l’expression des gènes de Map et Mbo, au niveau transcriptomique et protéomique lors de l’infection.

Le poste fait partie du projet AMITUB, soutenu par l’agence ANR. Le projet sera mené sous la direction de Yann Héchard et Ascel Samba au sein du laboratoire EBI (UMR CNRS 7267) de l’Université de Poitiers, en collaboration avec le laboratoire ISP (INRAE Nouzilly), le laboratoire LDAR24 (Coulounieix- Chamiers) et le LNR (Anses, Maisons-Alfort). Nos laboratoires ont une longue expérience dans les études sur les amibes et les mycobactéries.
Le candidat devra avoir une solide expérience des interactions hôte-pathogène, notamment via des approches transcriptomiques et/ou protéomiques. Des compétences en rédaction en anglais sont attendues.

Durée et salaire. Ce poste d’une durée de 2 ans (à temps plein) débutera idéalement en septembre 2024. Salaire net d’env. 2 300 euros par mois.

Post-doc position (two years): Molecular interactions of Mycobacterium bovis and Mycobacterium avium ssp. paratuberculosis with environmental amoebae (Poitiers, France)

Environmental free-living amoebae (FLA) have been hypothesized as reservoir components and vectors of Mycobacterium bovis (Mbo) and/or M. avium ssp. paratuberculosis (Map) and/or other mycobacteria that interfere with the diagnosis of tuberculosis. They could have an important role in the persistence of bovine tuberculosis and paratuberculosis respectively. Indeed, it has been experimentally shown that some mycobacteria may persist or multiply within FLA in vitro, but very few studies have addressed the role of environmental FLA in allowing survival and shaping virulence of mycobacteria. The objectives of the project are 1) to study the permissiveness of environmental FLA isolates to Mbo and Map infection in vitro and within mesocosms and 2) to decipher gene expression of Map and Mbo, at the transcriptomic and proteomic level during infection.

The position is part of the AMITUB project, supported by the ANR agency. It will be conducted under the supervision of Yann Héchard and Ascel Samba within the EBI laboratory (UMR CNRS 7267) at the University of Poitiers, in collaboration with the ISP laboratory (INRAE Nouzilly), the LDAR24 laboratory (Coulounieix-Chamiers) and the LNR laboratory (ANSES, Maisons-Alfort). Our labs have a long-term experience in amoeba and Mycobacterium studies.

The candidate should have a strong experience in host-pathogen interactions, especially via in transcriptomics and/or proteomics approaches. Skills in writing in English are expected.

Duration and salary. This 2-year (full-time) position will ideally start in September 2024. Net salary of ca. 2,300 euros per month.
